ค้นพบสูตร โดยคุณหมอชาวญี่ปุ่น ชื่อ Dr.Minoru Shirota ในปีคศ 1930 เนื่องจากต้องการรักษาผู้ป่วยของเค้าจากโรคระบบย่อยอาหาร คุณหมอได้พยายามวิจัยจากเชื้อแบคทีเรียมากมายในนมที่สามารถช่วยเพิ่มประสิทธิภาพในการย่อยของร่างกาย จนในที่สุดเจอเจ้าตัวนี้แหละชื่อ Lactobacillus casei Shirota Strain (ให้เกียรติโดยตั้งตามชื่อผู้ค้นพบ) ซึ่งเจ้าแบคทีเรียจิ๋วนี่จะอาศัยอยู่ในลำไส้ของเรา ช่วยปรับสมดุลแบคทีเรียที่มีอยู่ในลำไส้เรากว่า 100 ล้าน ล้าน ล้าน (ใช่ ล้านสามหนนั่นแหละ เค้าเรียกว่า trillion) ซึ่งบางชนิดมีประโยชน์ เช่น ตัวเจ้า Lactobacillus นี่แหละ ช่วยในเรื่องการดูดซึมสารอาหาร ทำให้ระบบย่อยมีประสิทธิภาพยิ่งขึ้น

ครอบครัวชิโรตะ ทำธุรกิจขายส่งกระดาษและเลี้ยงไหมจึงมีฐานะค่อนข้างดี แต่ชีวิตความเป็นอยู่ของคนในหมู่บ้านอินาดนิ บ้านเกิดของเขานั้นลำบากมาก แถมยังมีโรคระบาด ด้วยความที่เป็นคนหัวดีและเรียนเก่ง เขาตั้งใจจะเป็นแพทย์ตามความต้องการของพ่อ

ความแก่ชราของมุนษย์เกิดจากแบคทีเรียในลำไส้ถูกสารพิษทำลาย แต่ผู้คนแถบบัลแกเรียอายุยืนได้ก็เพราะกินโยเกิร์ตกันเป็นประจำ ซึ่งจุลินทรีย์แลคโตบาซิลลัสในโยเกิร์ตนี่แหละที่ช่วยกำจัดสารพิษในลำไส้ (ดร. เอลี เมชนิคอฟฟ์ นักชีววิทยา ชาวรัสเซีย)

แต่หลังจากนั้น จุลินทรีย์แลคโตบาซิลลัสก็จะตายเพราะกรดจากกระเพาะอาหารทำให้ไม่สามารถมีชีวิตรอดอยุ่ได้ในลำไส้ ชิโรตะคิดค้นวิธีเพื่อที่จะเลี้ยงเชื้อจุลินทรีย์เหล่านี้ให้มีชีวิตรอดในลำไส้ได้ โดยการพัฒนาสายพันธุ์ใหม่ที่แข็งแรงกว่า ซึ่งก็คือ สายพันธุ์ ชิโรต้า ตามชื่อของเขา

"ยาคูลท์" เป็นภาษาเอสเปอแรนโต มีความหมายเหมือนกับคำว่า "โยเกิร์ต" แปลว่า การมีอายุยืนยาว และที่หลายคนสงสัยว่าทำไมขวดเล็กแถมทำไมขวดต้องคอดเข้าแบบนี้ด้วย ก็เพราะว่าในปริมาณ 65 มล. จะมีจุลินทรีย์ยาคูลท์อยู่มากกว่า 15,000 ล้านตัว ซึ่งเพียงพอสำหรับการสร้างความสมดุลภายในลำไส้แล้ว ส่วนที่ขวดคอดก็เป็นการออกแบบมาเพื่อให้ถือสะดวก และ ลิ้มรสช้าๆ ไม่ดื่มทีเดียวหมด
