My PhD
I joined the Tissue Engineering team at University of sheffield in Oct 2005 as a PhD student and is currently researching the effects of mechanical stimulations on engineered bone engineered tissue. I was attracted to tissue engineering due to its interdisciplinary nature.
Background
Chiangmai University (Thailand) Doctor of dental surgery (DDS.)
Research
My PhD focuses on culturing bone forming cells and human mesenchymal stem cells(hMSC) on 3D scaffolds and trying to stimulate cell to produce bone by mechanical loading as well as studying the effects of primary cilia in bone cells.
Techniques used include:-
Dynamic compressive loading using Biodynamic chamber bioreactor (BOSE) Collagen staining Calcium staining Alkaline Phosphatase activity Immunostaining MTT/MTS assays, cells viability Realtime RT-PCR to monitor messenger RNA in the cells 3 dimensional scaffolds (Polyurethane, collagen etc.) Confocal microscopy Scanning electron microscopy (SEM) Oscillatory fluid flow parallel plate chamber SiRNA transfection Human mesenchymal stem cells (hMSC)
The challenges in this area include how the technology involved in developing mechanical loading of bone tissue constructs can be better developed to build new bone in the laboratory before implantation in human bone defects.
